Mattie's parents were both German Baptist Brethren and she was baptized when sixteen.  Mattie graduated from high school in 1899 and spent 1900 - 1903 in the Bible Department at Manchester College.  She and her brother, Joe Cunningham, were the first African American students to attend Manchester College.

In 1907 she married Newton Dolby and that same year she and Newton Dolby were installed as deacons in the Frankfort congregation.  On Dec. 30, 1911, Mattie Dolby was installed into the ministry in the Church of the Brethren.  She was one of the first, if not the first, Brethren women to be installed in the ministry.
